Project Experiences

1. Structural Biology Study of the T6SS-Secreted Esterase Effector Proteins in Pseudomonas aeruginosa [Project member (Master)]

  • Expression and purification of the core subunit VgrG2a of Pseudomonas aeruginosa H2-T6SS;
  • Optimization of negative staining for the VgrG1a & PA0260-PA0259 complex;
  • Sample preparation and Cryo-EM data collection for the VgrG1a & PA0260-PA0259 complex.

2.The structure analysis of the IBT-CoV144-Spike complex [Project main member (Master) ]

  • Sample Preparation with FEI Vitrobot Mark IV;
  • Using serialEM software collected Cryo-EM data;
  • Single particle analysis by Cryosparc and Relion.

3.Building a Molecular Docking and Molecular Dynamics Simulation Platform [Project main member (Research Assistant) ]

  • Installing Ubuntu 22.04.4 LTS on a Workstation;
  • Compiling and Installing GROMACS and Rosetta from Source code;
  • Through the established molecular dynamics simulation platform, the properties of various protein complexes were explored, such as the c-di-GMP and SodA complex, peptides and the SARS-CoV-2 N protein, and the spike protein and nanobody complex.
